چکیده

As is the case with diagnostic nuclear medicine, a major consideration in the development of targeted radiotherapeutics is to seek a balance between conceptual elegance and practicality. Nowhere is this more apparent than in the selection of the nature of the radiation to be exploited and the radionuclide that will be used to deliver this radiation. From a pragmatic perspective, convenient availability and distribution of the radionuclide at a reasonable cost are paramount. The multiday half-life b-emitters 131I and 90Y are notable examples and it is not coincidental that the first 2 Food and

Systemic Targeted Alpha Radiotherapy for Cancer

Background: The fundamental principles of internal targeted alpha therapy for cancer were established many decades ago.The high linear energy transfer (LET) of alpha radiation to the targeted cancer cellscauses double strand breaks in DNA. At the same time, the short range radiation spares adjacent normal tissues. This targeted approach complements conventional external beam radiotherapy and ch...

Radium-223 dichloride bone-targeted alpha particle therapy for hormone-refractory breast cancer metastatic to bone

BACKGROUND Hormone-refractory breast cancer metastatic to bone is a clinically challenging disease associated with high morbidity, poor prognosis, and impaired quality of life owing to pain and skeletal-related events. In a preclinical study using a mouse model of breast cancer and bone metastases, Ra-223 dichloride was incorporated into bone matrix and inhibited proliferation of breast cancer ...

The Promise of Targeted -Particle Therapy

The use of monoclonal antibodies to deliver radioisotopes directly to tumor cells has become a promising strategy to enhance the antitumor effects of native antibodies. Since the and -particles emitted during the decay of radioisotopes differ in significant ways, proper selection of isotope and antibody combinations is crucial to making radioimmunotherapy a standard therapeutic modality. Becaus...

Enhancing and verification of dose in external radiation therapy using Gd nanoparticles as a theranostic agent: A Monte Carlo simulation study

Introduction: Theranostics, in particular, the use of radionuclides with the capability of simultaneous imaging and treatment has opened new horizons in personalized treatment planning of targeted radiation therapy. In this approach, positive beta or gamma emitters are required for imaging and alpha, beta and Auger electrons for treatment purpose. On the other hand, studies hav...

Targeting Prostate Cancer Stem Cells with Alpha-Particle Therapy

Modern molecular and radiopharmaceutical development has brought the promise of tumor-selective delivery of antibody-drug conjugates to tumor cells for the diagnosis and treatment of primary and disseminated tumor disease. The classical mode of discourse regarding targeted therapy has been that the antigen targeted must be highly and homogenously expressed in the tumor cell population, and at t...
